Ok I haven't been in over 20 years. Growing up I think it was mostly field trips when I visited and I remember always hitting the tram ride first....
What are suggestions for Universal Studios today? I know Harry Potter is now open. Anything to make sure to hit first?
Is there anything there that deal with special effects makeup, either stores or exhibits?

I went this summer and really enjoyed it. We did the VIP experience, so I can't really speak much to the order of things. I did observe that the Harry Potter area was much more packed in the afternoon and evening than it was in the morning when we visited it.
I don't remember anything specifically with special effects makeup. There is a show in FL, but I know I didn't see it in Hollywood, if it is there also. The "scareactors" at Halloween Horror Night have serious special effects makeup, if you are there in the next few weeks.

DH & DS went in May to see HP. We live in Seattle, but found an annual pass was a good deal and it allowed them early entry to HP too. I haven't checked recently but when they went in May, HP was least crowded in the morning and evening and packed mid-afternoon (so avoid mid-afternoon).
I think it is really a 1 day park but DH and DS went 2 days and took there time, rode rides and the tram tour twice or more.
The Disboards has a California section with good info on USH.

We went in April - 2 weeks after Harry Potter opened.
We went on an early opening morning (Saturday), and were at the gates at 7am when they opened. While it was insanely early for us, it was perfect. We walked onto the main ride with no wait, and did everything else in that section before 8:30am. We went to go back in early afternoon and it was PACKED.
Next we did the tram, then the other rides. We did pretty much everything there, stood in lines for all the photo ops, and saw the shows before 7pm.
It was very much worth it to go early and do HP first.

I debated about the horror nights (we will be there in a week) but think I need to put that off a couple more years. Daughter will be turning 13 on Halloween and has never been to Un.St. So this will be a good fun visit! She's been wanting to do a haunted house but not knowing how she will react I'm not taking a chance! She been playing around with making herself into a zombie for Halloween with store bought makeup and concoctions she is making out of household material.

Thank you. Looks like what I thought is to hit HP first and we can revisit later if needed. We will be going on a Friday and hopefully schools down here will not be off. I think hours are 9-6 with early admission at 8 (I think if you buy online? Need to look into that).

We always went right to the tram first whenever we visited so that we didn't get caught in lines later, but I'm sure the best thing to do is hit HP first right now. I have to put in a plug for the Waterworld show for stunts and special effects. There will only be a couple a day and they will be listed on a "what's happening today" sheet that you can get when you go in to the park (this sheet will also tell you the last tram ride time which is at least an hour before the park closes). DD and friends bought season passes when HP opened and always go on Fridays. They don't even get to the park until after noon and haven't run into any significant lines. I don't think schools have a day off until Nov 11, so you've picked a great time to visit.

My daughter is off Friday so pulling her out of electives on Wednesday... Normally off Thursday. Usually we are down here over the holidays and I refuse to go then. I do remember a live show we would always catch so will make sure to look for the schedule of events to hep plan the day.
