If I want to feed 12" vinyl directly into the machine I know I have to adjust the rollers, but do I need to adjust my image location on the computer too? I mean do I have to shift the image to the left because the media is no longer centered on a cutting mat? I'm cutting a large image that is 11 1/2" wide. Would it be better to use the mat instead? I don't want to cut it off and waste vinyl.

If you are loading the media directly into the Cameo, you need to have it set to "Load media" instead of "Load cutting mat" (or whatever it says).
However, I have never actually done it. I have always used my mat and just cut the vinyl. Yes, it hurts to waste anything, but if you pay attention to how your design is placed on the mat, you can cut and place the vinyl in a way that minimizes the waste.
One reason I prefer to use the mat is that if I were to forget to adjust my blade, or it was not set right in some way, you can damage the strip underneath, and that can cause errors for future cutting. If it cuts into the mat, the mat is easy to replace. The strip underneath has to be ordered online.
I still save any scraps for smaller vinyl designs/cuts, which can often be used up for layered designs. Just be sure you trim off any excess before you start to weed.
Also, if you are using HTV (heat transfer vinyl), remember to mirror your image before cutting.
