How long to make it worthwhile depends on how far, who you are seeing and how you feel about staying with them ect ... how far are you from Florida? From NY to Oklahoma (driving 30+ hours with three kids)then staying with my parents I felt a month was worthwhile. From Oklahoma to Missouri, 3 days is a good trip length.

We drive from Chicago to Florida every year. Leave at 5pm and get there around 11am the next day. Dh naps during the day, drives from 5p-5a and then I take over and he naps. We usually go straight to Disney and spend the day. We are a little tired. Kids sleep a full 8 hours so they are fine. We feel like we don't miss any time and it's better and cheaper than flying. We usually spend 7-8 days in Florida and head back over night again.

I have done a 24 hour drive a couple times. I don't sleep well in the car so the first day there and the first day home are always a challenge for me.

at least 4-5 days (and that's added to the 2 days on either end for driving 1000 miles). But if we can get some place in 1 day, then we've been known to stay only 2-3 days. We don't drive overnight anymore, but prefer to stick to less than 10 hours each day. Just the other day though we drove 13 the first day and 5 the second (trying to make it at a specific time the 2nd day to make a HS graduation ceremony with plenty of time to spare).

The time we spend somewhere would depend partly on where that 'somewhere' is. Where in Florida will your parents live? I could spend more time there if it was on the beach or near Orlando. We visit Florida every year, and even though we obviously don't drive there (there's no Atlantic Tunnel yet!), it takes us approximately 18 hours door-to-door. I wouldn't do that journey for any less than ten days.

For long road trips, I look at your question the other way -- is there enough at the staying point to make the drive worthwhile.
