I plan to drive up 55 to Chicago from St Louis, and I want to stay in a hotel where I can use public transportation to get into the city in about 30-45 minutes. What area should I be looking in?

Maybe Oakbrook area. I'm not totally sure about the train schedule.
If you find a hotel, then ask them about the train.

I think anything on a map that is a near western or near southwestern suburb. Do you want to park and ride or walk to the station? Lombard has several hotels and a station, but I don't know if any are walking distance. I don't know if Oak Brook has a station, but they have hotels. I think you could also look by lemont, which is down by 55. I don't know hotels down there though.
ETA. If you are staying outside the city to avoid driving there, think about oak park. It's too much for me.

Oak lawn hilton is close to train line and not far off 55. Train schedule is more limited than other lines.
Metrarail.com will show train schedules for all their lines.

Lombard has a station, but no hotels within walking distance. However, there are commuter lots right there and you can pay to park for the day. It's quite reasonable. It has a nice, but small downtown area, too, with some good restaurants. There are hotels nearby, including a nice Crowne Plaza (at the corner of Finley & Roosevelt, in case you search) with one of my favorite restaurants in it (Glen Prairie). It's about 2.5 miles from the train station.
Downers Grove would be another option; again, no hotels within walking distance, but a commuter lot.
ETA: I wouldn't stay near Midway, but if you do, you'd take the orange CTA line in. You will have to buy a Ventra card.
You could also stay in Rosemont and take the Blue Line. Rosemont has a ton of hotels, most of which will shuttle you over to the blue line stop, and a ton of entertainment right there in that big complex.

LaGrange has a really nice downtown area and some great restaurants. I know there is public transportation there (metra), just not sure where it is in LaGrange. We have friends that live there and their kids took the train into the city to go to private high school. Great little town.
I wouldn't stay near Midway - enjoy a smaller town. Around Midway is being in the city and that area has nothing endearing about it.
Tinley Park is right off I-80 (you can take 55 to 80) and there are two train stations and a few hotels off of Harlem. You can park at the train stations, but most trains take 45-60 to get downtown.

Yes, it is a nice town (I used to work there), but I don't think there are any hotels. SockMonkey: You can still buy single-use disposable CTA tickets. It's convenient for a seldom-user, but a little bit of a rip off at $3 a ride: $2.25 for the fare, 25¢ for a transfer, and 50¢ as punishment for not buying a Ventra card.
